A comparative study of different extracts of Coptis chinensis on type 2 diabetic rats
OBJECTIVE To compare the contents of effective components in different extracts of Coptis chinensis and their therapeutic effects on type 2 diabetic rats, and to explore the correlation between the contents of main components and their therapeutic effects. METHODS: The contents of representative active ingredients in supercritical fluid extract of Coptis chinensis and simulated gastric juice extract of Coptis chinensis were determined by high performance liquid chromatography (HPLC). Ninety adult male Wistar rats were selected as subjects, and 15 rats were randomly selected as normal control group (group N) and fed with standard rat diet. The rest rats were fed with high fat diet and injected with small dose of streptozotocin (STZ) into tail vein to establish 2. Diabetic rats were randomly divided into model control group (M group), positive control group (D group), Coptis supercritical extract treatment group (A group) and Coptis simulated gastric juice extract treatment group (B group). After 9 weeks of continuous treatment, the body weight, OGTT blood sugar level, serum triglyceride (TG) and interleukin-6 (IL-6) levels and pathological status of intestinal tissues were compared. RESULTS: (1) At the same dosage, the contents of the representative effective components in the supercritical extracts of Coptis chinensis were higher than those in the simulated gastric juice extracts of Coptis chinensis; at the effective dosage, the contents of berberine and palmatine in the supercritical extracts of Coptis chinensis were only higher than those in the simulated gastric juice extracts of Coptis chinensis; and (2) After 9 weeks of treatment with the effective dosage, OGTT blood sugar level, body weight and body weight of rats in groups D, A and B were increased. Serum levels of TG and IL-6 were significantly lower than those of group M (P < 0.05); there was no significant difference between group A and group B (P > 0.05); under light microscopy, HE staining of intestinal tissue in group D, group A and group B showed that inflammatory improvement was significantly better than that in group M. ConclusionCoptis supercritical extract and Coptis simulated gastric juice extract have a certain therapeutic effect on type 2 diabetic rats. They can significantly reduce blood sugar, blood lipid, body weight and serum IL-6 levels, and improve chronic intestinal inflammation. The content of the main components has no correlation with the curative effect.
